Scleroderma patients derived fibroblast have been isolated and cultured for the drug testing. More than 500 drugs are now being tested on those cells which takes quite a long time. Several fibroblast markers have been used for characterisation.

The aim of the project was to develop 'Gene therapy for APECED patients', by delivering correct version of 'AIRE' gene in mutated cells. I started with the APECED disease model rats, genetically modified by Pärt Peterson, University of Tartu. It was a kind gift from Pärt that facilitated us to isolate the Rat epidermal Keratinocyte cells (REK). REK cells were immortalised with commercial SV40 and hTERT Lenti viruses. Immortalised cells have been infected with Endo Aire Lenti virus to correct the mutation with puromycin selection gene. Next plan is to do single cells RNA seq analysis and delivering the corrected cells to Aire -/- rats at Tartu, Estonia.

Project: 'Adipocyte differentiation from MSCs cells'

My thesis title was 'Adipocyte differentiation from hESCs derived MSCs, comparing two different protocols.' Following two protocols, I successfully differentiated Mesenchymal stem cells from hESCs. Then adipocytes were differentiated from MSCs. Mesenchymal progenitor cells were characterised by flow cytometry analysis, immunostaining and qPCR analysis and adipocytes were evaluated by oil red o staining and qPCR. Later I also explored IL1 alpha, beta, and IL receptor antagonist expression level in different stages of mesenchymal stem cell differentiation. Furthermore, I induced autophagy in MSCs with different concentrations of rapamycin to see if autophagy affects the IL1beta secretion level. Moreover, I also performed a lot of western blot analysis and confocal imaging of different stages of mesenchymal cells. I also collected the supernatant to apply ELISA technique for sensitive analysis of IL1b secretion.
